Act as cloning vector, Able to replicate freely in the host cell (Ori), Able to accept foreign DNA in multiple cloning sites (MCS), Having selectable marker gene/ genetic marker (ampR), The pUC18/ plasmid/ lacZ gene will be recognized and cut/cleaved by EcoRI at 5’ GAATTC 3’ , Produce compatible/ complementary sticky ends, This helps to identify/ select for bacteria that have taken up/ acquired pUC18/ the plasmid, Only bacteria with/ that have taken up pUC18/ the plasmid will be able to grow in the presence of antibiotic ampicillin.
